Для синтеза этого фермента использовали S30-клеточный экстракт Escherichia coli, химерную РНК-полимеразу бактериофага Т7 и высококопийный плазмидный вектор pET42mut со встроенным в него геном браззеина. В результате выполнения работы впервые продемонстрирована возможность синтеза сладкого белка в системе БСБ. При этом объемный выход браззеина в оптимизированных условиях проведения процесса составил 2 мг/мл реакционой смеси, что в 57 раз больше, чем максимальный выход его в ранее описанных в литературе экспериментах с использованием цельноклеточных систем экспрессии.</p></abstract><trans-abstract xml:lang="en"><p>Feasibility of producing brazzein plant sweetener by the cell-free biosynthesis procedure as an alternative to the classical submerged fermentation method was assessed in the present investigation. Chimeric RNA polymerase of T7 bacteriophage, S30-cell extract of Escherichia coli and multicopy plasmid vector pET42mut with the inserted brazzein gene were included into the protein synthesis. The completed research resulted in the first successful demonstration of the sweet protein biosynthesis in the cell-free system.
